Aim of study (acc. to author)

The effects of exposure of induced pluripotent stem cells to a pulsed 50 Hz magnetic field on osteogenic differentiation should be investigated.

Cells were divided into 4 groups: 1) exposure to the magnetic field, 2) treatment with osteogenic medium (3 mM beta-glycerophosphate, 50 µg/ml ascorbic acid and 10-9 M dexamethasone), 3) co-exposure to the magnetic field and osteogenic medium, 4) control group.

After 5 and 7 days, all exposure groups (groups 1-3) showed a significantly increased proliferation compared to the control group. However, the co-exposure group (group 3) showed the highest value, respectively, which was even significantly higher than in groups 1 and 2.
Mineralization of the extracellular matrix was most distinct in group 3 as well and the calcium content, enzyme activity of alkaline phosphatase and the gene expression of most osteogenesis-related genes (collagen type 1, osteocalcin, alkaline phosphatase, osteoprotegerin, matrix metalloproteinases 1 and 3) were also most distinct in group 3 and significantly increased after 7 and/or 14 days compared to groups 1 and 2.
The authors conclude that exposure to a pulsed 50 Hz magnetic field in combination with osteogenic medium might be beneficial for osteogenic differentiation of induced pluripotent stem cells. These findings may be of therapeutical use.
